Arginase inhibitor INCB-1158 restores Arg level in sera and TIFs in syngeneic MC38 mouse colorectal cancer model.C57BL/6J mice were subcutaneously inoculated with MC38 colorectal cancer cell line and treated with 100mg/kg of INCB-1158 for one or five consecutive days. Sera (A) and TIFs (B) were collected after each treatment period and subjected to Arg level quantification using #IS-I-0400R Arg ELISA kit.
Cross-validation of Arginine ELISA and LC/MS data in human plasma samplesArg was quantified in plasma samples from 40 human subjects either using IS-I-0400 ELISA kit or by liquid chromatography–mass spectrometry (LC/MS). Correlation coefficient of 0.9713 confirms the accuracy of the immunoassay.
Typical standard curve of Arginine ELISATypical standard curve obtained with the Arg ELISA kit. In this competitive enzyme immunoassay, optical density is inversely correlated with hArg levels within a linear range of 5.12 - 200µM. (example data - do not use for calculation).
Arg metabolism is predictive of anti-PD(L)1 immunotherapy-treated patient outcomeQuantification of baseline plasmatic Arg in non-durable clinical benefit (NDB ; n= 214, green) and durable clinical benefit (DCB ; n= 80, yellow) patients. p value was calculated using Wilcoxon test.
Arg metabolism is predictive of anti-PD(L)1 immunotherapy-treated patient outcome(A) Rate of clinical benefit (NDB; Non-durable vs DCB, durable clinical benefit) of advanced cancer patients treated with ICB according to baseline plasmatic Arg level (Low vs High). p-value is calculated using Khi-deux test. (B) Kaplan Meier curves of progression-free survival according to baseline plasmatic Arg levels.

Yes, this kit allows to quantify arginine in cell culture supernatants.
The Arginine (L-Arg) ELISA kit enables the quantitative determination of the non-essential amino acid L-Arginine in serum, plasma, and cell culture supernatants. Validated in human plasma with a high correlation to LC-MS results, the assay has also been successfully applied to mouse serum samples (see supporting data). Designed for versatility, the kit is species-independent and requires only a small sample volume (20 µL), making it well-suited for both preclinical research and clinical studies.
